Hi, I’ve never had any bees in a house before but we raised bees on the farm where i grew up. Blocking the hole is not going to help as the bees are most likely going to find another route. As far as the numbers of bees go ,its most likely in the thousands. Bees usually build vertically and cap their brood and honey. Is it possible the wet spots in your ceiling are caused by another reason?Have you thought of consulting a local bee keeper to harvest the bees from your home? Honeybees are having a bad time right now and there may be someone close by that would be happy to get them, if not You should consult an exterminator about ridding your house of the bees. Getting rid of the bees first might be best ,as working on your ceiling could possibly breach the hive. We transfered a couple of wild colonies from bee trees years ago and bees tend to get upset when their home is opened up. Good luck with your unique problem. Here is a link to a page that might help.
